An Engineered Heat-Inducible Expression System for the Production of Casbene in Nicotiana benthamiana

چکیده

Plants respond to heat stress by producing heat-shock proteins. These are regulated promoters containing regulatory elements, which can be harnessed control protein expression both temporally and spatially. In this study, we designed heat-inducible produce the diterpene casbene in Nicotiana benthamiana, through a multi-step metabolic pathway. To potentially increase gene transcription, coupled elements from Arabidopsis thaliana Hsp101 or Glycine max GmHsp17.3-B promoters, CAAT TATA boxes CaMV 35S, 5′UTR tobacco mosaic virus. The resulting four chimeric fused green fluorescent (GFP) reporter showed that variant Ara2 had strongest signal after shock. We next created 4-gene cassette driven promoter allow for exogenous synthesis of transformed multigene construct along with selectable marker into benthamiana. Metabolic analysis on transgenic lines revealed continuous outperforms shock, up 1 μg/mg DW detected 32 h uninterrupted 40 °C heat. results demonstrate potential as synthetic biology tools metabolite production plants.

Production of an engineered killer peptide in Nicotiana benthamiana by using a potato virus X expression system.

The decapeptide killer peptide (KP) derived from the sequence of a single-chain, anti-idiotypic antibody acting as a functional internal image of a microbicidal, broad-spectrum yeast killer toxin (KT) was shown to exert a strong microbicidal activity against human pathogens. With the aim to exploit this peptide to confer resistance to plant pathogens, we assayed its antimicrobial activity again...

Optimization of Engineered Production of the Glucoraphanin Precursor Dihomomethionine in Nicotiana benthamiana

Glucosinolates are natural products characteristic of the Brassicales order, which include vegetables such as cabbages and the model plant Arabidopsis thaliana. Glucoraphanin is the major glucosinolate in broccoli and associated with the health-promoting effects of broccoli consumption. Toward our goal of creating a rich source of glucoraphanin for dietary supplements, we have previously report...

The production of human glucocerebrosidase in glyco‐engineered Nicotiana benthamiana plants

For the production of therapeutic proteins in plants, the presence of β1,2-xylose and core α1,3-fucose on plants' N-glycan structures has been debated for their antigenic activity. In this study, RNA interference (RNAi) technology was used to down-regulate the endogenous N-acetylglucosaminyltransferase I (GNTI) expression in Nicotiana benthamiana. One glyco-engineered line (NbGNTI-RNAi) showed ...

Techno-economic analysis of horseradish peroxidase production using a transient expression system in Nicotiana benthamiana.

Despite the advantages of plant-based transient expression systems relative to microbial or mammalian cell systems, the commercial production of recombinant proteins using plants has not yet been achieved to any significant extent. One of the challenges has been the lack of published data on the costs of manufacture for products other than biopharmaceuticals. In this study, we report on the tec...
